The aim of the study was to establish a highly efficient, simple method to extract and purify soil microbial DNA. The extraction of microbial DNA fragment was about 23 kb in size, the yield of DNA solution after purification was（7.76 ±1.53）μg·g^-1, OD₂₆₀/OD₂₈₀ was 1.78 ±0.04, OD₂₆₀/OD₂₃₀ was 1.76 ±0.02.
