Soybean cyst nematode is devastating pathogens that infect soybean and can cause 10%~20% loss ofyield. Plant-host mediated RNAi is one of effective strategy to breeding new insect-resistant transgenic plants. Hgrps-23, which encodes ribosome protein of SCN was cloned into RNAi vector. Then, RNAi construct were trans-formed into Williams82 by Agrobacterium tumefaciens-mediated transformation. Analysis of transgenic plants usingPCR and Southern blot method has proved that the Hg-rps-23 RNAi fragments have inserted into the soybean ge-nome. T2 transgenic plants were grown in soil with cysts and the results showed that the transgenic plants’ resis-tance to SCN3 was significantly higher than that of receptor soybean. Our study suggested that plant- mediatedRNAi could enhance the resistance of soybean to SCN. It also provides a new approach to generate SCN-resistantsoybean materials and new cultivars.
