    摘要:

    从健康人参根际土壤中分离获得一株生防菌株ATB4。根据16S rDNA、gyrA基因序列分析,结合形态学、生理与生化特性鉴定,将生防菌株ATB4鉴定为甲基营养型芽孢杆菌(Bacillus methylotrophicus)。通过单因素试验和正交试验设计,对其发酵培养基成分培养条件进行优化,探究生防菌对引起人参锈腐病的Cylindrocarpon destructans防治效果,最终得到该菌最适培养基成分为3%葡萄糖、4%酵母浸粉、0.05%MnSO4·7H2O和0.1%K2HPO4,最佳发酵条件为4%接菌量,初始pH 8.0、培养温度28℃、培养24 h、250 mL三角瓶装液量40 mL。

    Abstract:

    One antagonistic bacterial strain ATB4 was isolated from the rhizosphere soil samples of healthy ginseng.Based on phylogenetic analyses of 16S rDNA and gyrA gene sequences, and morphological, physiological, biochemical characterizations, the strain ATB4 was identified as Bacillus methylotrophicus. The single factor experiment and orthogonal experiment were adopted to optimize the liquid fermentation medium and conditions that enhance its bio-control efficacy against Cylindrocarpon destructans caused ginseng rusted root. The best compositions of cultural medium were as follow: 3% glucose, 4% yeast extract powder, 0.05% Mn SO4·7H2O and 0.1% K2HPO4. The optimal fermentation conditions were as follow: inoculation rate 4%, initial pH 8.0, temperature 28℃, fermentation time 24 h,and medium capacity 40 mL/250 mL.
