    以铁皮石斛幼嫩的带节茎段为外植体,通过研究植物生长调节剂对类原球茎诱导、增殖和分化的影响,筛选出适宜铁皮石斛类原球茎植株高效再生的条件。结果表明:类原球茎诱导的最适培养基为MS+6-BA 1.0 mg/L+IBA 1.0 mg/L+土豆泥60 g/L+活性炭0.5 g/L+蔗糖25 g/L+琼脂6.5 g/L,茎段接种45 d后,类原球茎诱导率为97.78%;类原球茎增殖和分化的最适培养基为MS+6-BA 2.5 mg/L+IBA 1.0 mg/L,培养30 d,增殖倍数为7.94,培养至60 d,丛生芽诱导率为97.33%;再生苗在MS+NAA 0.5 mg/L培养基中培养30 d,生根率达到100%。

    Abstract:

    In order to establish an efficient plant regeneration system,young stem segments of Dendrobium officinale Kimura et Migo were used as explants,and the effects of plant growth regulators on the protocorm-like body(PLB) induction,proliferation and differentiation were studied. The results showed that the optimum medium for PLB induction was MS + 6-BA 1.0 mg/L + IBA 1.0 mg/L + mashed potato 60 g/L + acticarbon 0.5 g/L + sucrose 25 g/L + agar 6.5 g/L,and the PLB induction rate was 97.78% after cultured for 45 d. The optimum medium for PLB proliferation and differentiation was MS + 6-BA 2.5 mg/L + IBA 1.0 mg/L,the proliferation multiple of PLB reached 7.94 after cultured for 30 d,and the induction rate of cluster bud was 97.33% after cultured for 60 d. The optimum rooting medium was MS + NAA 0.5 mg/L,and the rooting rate was 100% after cultured for 30 d.
