    摘要:

    为了提高枯草芽胞杆菌GB519发酵液菌体数量和芽胞率的统计效率,本研究采用平板涂布法和平板倾注法进行活菌计数,利用加热除菌法、结晶紫染色法和孔雀绿染色法开展芽胞率检测效率的比较研究。结果表明:平板涂布法与倾注法的菌体计数数量差异不大,误差率相对一致。枯草芽胞杆菌GB519利用结晶紫染色法与孔雀绿染色法检测芽胞率差异不显著。确定了枯草芽胞杆菌GB519加热除菌法菌体致死温度为80℃、时间为15 min,其芽胞量为131.9×108CFU/mL,芽胞率为50.1%,与孔雀绿染色法(54.8%)差异不显著,与结晶紫染色法芽胞率(58.7%)差异显著。本研究将为类似的芽胞检测研究提供方法和数据参考。

    Abstract:

    In order to improve the statistical efficiency of the sporular amount and germination rate in the fermentation broth of Bacillus subtilis GB519,the methods both plate coating and plate pouring were used to count the viable bacteria. And the methods of heat sterilization,crystal violet and malachite green staining were used to compare with the germination rate. The results showed that there was little difference in sporular amount between plate coating and pouring method,and the error rate was relatively consistent. It was determined that the lethal temperature and time of heat sterilization method were at 80℃ for 15 min,and the sporular amount and germination rate of that were 131.9×108 CFU/mL and 50.1%,respectively,which was no significantly different from malachite green staining(54.8%),and was significantly different from that of crystal violet staining(58.7%). This study could provide positive conferences from both parameters and methods for the similar research.
