In order to improve the statistical efficiency of the sporular amount and germination rate in the fermentation broth of Bacillus subtilis GB519,the methods both plate coating and plate pouring were used to count the viable bacteria. And the methods of heat sterilization,crystal violet and malachite green staining were used to compare with the germination rate. The results showed that there was little difference in sporular amount between plate coating and pouring method,and the error rate was relatively consistent. It was determined that the lethal temperature and time of heat sterilization method were at 80℃ for 15 min,and the sporular amount and germination rate of that were 131.9×108 CFU/mL and 50.1%,respectively,which was no significantly different from malachite green staining(54.8%),and was significantly different from that of crystal violet staining(58.7%). This study could provide positive conferences from both parameters and methods for the similar research.
