Abstract:Based on the optimization of pretreatment conditions and chromatographic separation, a Highperformance liquid chromatography HPLC method for the determination of cannabidiol(CBD) and Δ9-tetrahydrocannabinol(THC) in industrial hemp was developed. The samples were extracted with 95% ethanol, acetonitrilewater(68∶32, v/v) as the mobile phase, and separated by C18 column at 35 °C with a flow rate of 1.0 mL/min, the injection volume was 10 μL and the detection wavelength was 220 nm. The results show that the method has a good linear relationship in the range of 0.02-0.10 mg/mL, and the correlation coefficients(r) are all greater than 0.998.The detection limits(LOD, S/N=3) and quantitative limits(LOQ, S/N=10) were 0.0001 g/100 g and 0.0003 g/100 g for CBD, 0.0006 g/100 g and 0.0018 g/100 g for THC, respectively. The recovery rates of CBD and THC were 92%-101% and 80%-102%, respectively when the negative samples were added with 0.5 mg/kg, 50 mg/kg and 200mg/kg. The method has good accuracy, sensitivity and stability, and can meet the detection requirements of CBD and THC in industrial hemp.
